Allgemeines Forschungsinteresse
Lee Spencers Interesse gilt der känozoischen Säugetierbiochronologie, der mesozoischen Taphonomie und der Paläobiologie sowie der Modellierung der Sintflut. Er arbeitet an einem Lehrbuch für vergleichende Wirbeltieranatomie und ist Mitglied in einer Arbeitsgruppe, die an der historischen Geologie Nordamerikas von einem kreationistischen Standpunkt aus arbeitet.

Gegenwärtige Forschung
Die gegenwärtigen Forschungsaktivitäten beinhalten die Taphonomie von spätkreidezeitlichen Knochenlagerstätten in der Lance Formation und der Morphologie von Dinosauriereierschalen in Argentinien.
